AAV capsid multi-property correlation
AAV capsid / expression cassette for a defined tissue
Across 10,000 AAV9 7mer variants, what fraction ranks at or above the 90th percentile in target tissue while at or below the 10th percentile in liver?
THE QUESTION
For a named rare-disease target and delivery route, which AAV capsid–expression-cassette combination achieves at least a 3-fold improvement in target-tissue transduction versus the chosen reference capsid while keeping off-target liver signal and neutralising-antibody escape within pre-specified limits?
WORK ORDER
Target-tissue versus liver tropism correlation across a shared 10,000-variant AAV9 7mer library
BACKGROUND
What this Apodex Lab is
Adeno-associated viral vectors deliver therapeutic genes to target organs, but systemic administration often deposits most of the viral dose in the liver. This off-target accumulation restricts the dose that can safely reach diseased heart, brain, lung, kidney, or muscle tissue, while raising the risk of hepatotoxicity and immune clearance. Capsid engineering modifies viral surface loops so vectors accumulate selectively in target tissues while sparing the liver. Gene-therapy programmes design engineered capsids under the assumption that high target delivery and low liver uptake can coexist in a single viral coat, yet vectors that satisfy both criteria simultaneously remain difficult to isolate.
The unresolved question is whether target-tissue tropism and liver accumulation are independent properties or positively correlated across sequence space. If variants that transduce peripheral organs also enter liver tissue more effectively, capsids that rank in the top decile for target transduction and the bottom decile for liver signal may not exist. Existing datasets do not settle this question because research groups have evaluated mouse tissue tropism, primate liver uptake, packaging viability, and major histocompatibility complex presentation in separate studies on unrelated variant libraries. Without measuring these properties on a single shared library, the correlation between non-liver tissue enrichment and liver de-targeting cannot be calculated.
The AAV capsid / expression cassette for a defined tissue Apodex Lab resolves this coupling by evaluating a shared library of 10,000 AAV9 variants with seven-residue insertions in the VP1 surface loop. The variants carry a constant reporter genome and are dosed intravenously in C57BL/6J mice. Vector-genome abundance across seven solid organs and serum is measured 21 days later by indexed amplicon sequencing to determine the Spearman rank correlation between maximum non-liver tissue enrichment and liver accumulation. If the correlation exceeds 0.5 and fewer than 0.5% of variants meet the dual-constraint threshold, developers can halt single-capsid multi-constraint screening campaigns and redirect liver de-targeting to expression-cassette engineering.
WHY NOW
Why the field is spending on an assumption
Engineered AAV development assumes that capsids can achieve high target-tissue transduction while detargeting the liver. Published selections measure mouse multi-organ tropism and macaque liver enrichment on separate libraries of 50,000, 200,000, and 63,000 variants, leaving the correlation between target and liver uptake unmeasured. Pooled multi-organ selection with indexed sequencing now allows multiple tissues to be quantified from a single shared library. Without this joint distribution, programmes design vectors around simultaneous high-target and low-liver constraints without direct evidence that any variant satisfies both.
WHAT WE’D BUY
What we would ask a laboratory to run
We are not buying the answer. We are buying a measurement with an interval — and the bars it will be compared against.
A gene-therapy contract research organisation packages a 10,000-variant library of seven-residue peptide inserts into AAV9 capsids with a CAG-GFP genome. After intravenous dosing in C57BL/6J mice, amplicon sequencing measures vector-genome DNA across seven tissues and serum at day 21. The measurement returns the Spearman rank correlation coefficient between maximum non-liver tissue enrichment and liver enrichment, bounded by a bootstrap 95% confidence interval no wider than 0.15. It also reports the fraction of variants simultaneously at or above the 90th percentile in a non-liver tissue and at or below the 10th percentile in liver, alongside concordance data from an independent 96-variant confirmatory pool.
SCOPE & LIMITS
Vector-genome abundance in healthy male C57BL/6J mice measures DNA accumulation rather than payload expression, and rankings derive from pooled intra-animal competition within a designed library rather than individual administrations.
